R570 is a modern sugarcane cultivar. Modern sugarcane cultivars are derived from a few interspecific hybridizations performed a century ago between the sugar rich domesticated species Saccharum officinarum (2n=8x=80) and the wild species Saccharum spontaneum (2n=40 to 128). R570 cultivar was obtained by CERF (now eRcane) in Réunion island (http://www.ercane.re/). The genome of this cultivar has about 115 chromosomes, including 10% of whole chromosomes derived from S. spontaneum, 10% from S. officinarum/S. spontaneum recombinant chromosomes, with the remaining being whole chromosomes from S. officinarum (D'HONT et al. 1996). Modern sugarcane cultivars have a total genome size of about 10 Gb while the monoploid genome size is about 800-900 Mb. The monoploid reference genome sequence was obtained by sequencing a mimimum tiling path (MTP) of 4660 BACs selected based on their alignment on the sorghum genome. The overlapping regions between BAC sequences were then trimmed to construct a single tiling path (STP) that is single copy in sugarcane. This strategy was deployed to overcome assembly difficulties linked to the high ploidy, aneuploidy and heterozygosity of sugarcane cultivars.

This sugarcane reference genome was obtained in the framework of an international collaboration coordinated by Cirad (Centre de Coopération Internationale en Recherche Agronomique pour le Développement) and involving JGI (Joint Genome Institute), Keygene, QAAFI (Queensland Alliance for Agriculture and Food Innovation), CSIRO (Commonwealth Scientific and Industrial Research Organisation), SASRI (South African Sugarcane Research Institute), USP (Universidade de Sao Paulo,) and financially supported by the International Consortium for Sugarcane Biotechnology (ICSB) and the Joint BioEnergy Institute (JBEI).
